چکیده

e principal function of the mammalian immune system is to combat infectious diseases. Immune responses to antigens and microbial pathogens are divided into those mediated by cells (i.e., cell-mediated immunity) and those mediated by antibodies (i.e., humoral immunity). e T lymphocyte, the “central player” in cell-mediated immunity, provides antigen speci€city and orchestrates the antimicrobial activities of cells that generally lack antigen speci€city. “Supporting players” in cell-mediated immune responses include dendritic cells (DCs), which present antigens to T cells and provide contextual information, and monocytes and macrophages, which carry out instructions provided by activated T cells. Antigen speci€city is conferred by T cells, which bear T-cell receptors (TCRs) responsible for the detection of pathogen-derived peptides presented on major histocompatibility complex (MHC) molecules. Primary encounter of naïve T cells with a pathogen results in their activation, proliferation, and di‚erentiation into e‚ector T cells. Aƒer pathogen clearance, e‚ector T cells contract and give rise to long-term memory T-cell populations. Our understanding of T-cell responses to microbial pathogens and of the complexity of cell-mediated immune responses have increased dramatically. e discussion in this chapter is focused on our current knowledge regarding T-cell responses to microbial pathogens.

Immunity to pathogenic free-living amoebae: role of cell-mediated immunity.

The role of cell-mediated immunity in defense against pathogenic free-living amoebae was examined. Both the in vitro macrophage inhibition test and the in vivo delayed hypersensitivity test showed responses to both heterologous and homologous antigens, although homologous systems were the most efficient. It is suggested that exposure to nonpathogenic species of free-living amoebae can stimulate...

Differential sensitivities of Chlamydia trachomatis strains to inhibitory effects of gamma interferon.

Gamma interferon (IFN-gamma) is an important cytokine in host defense against chlamydial infection. An in vitro cell culture system was used to show that IFN-gamma inhibition of chlamydial growth, as determined by diminished recovery of infectious elementary bodies, differed markedly among chlamydial strains. These differences in sensitivity among chlamydial strains to IFN-gamma-mediated inhibi...

Role of neutrophils in protection of udder from infection in high yielding dairy cows

Protection of the mammary gland against mastitis-causing pathogens is mediated by many factors in thegland and blood circulation. The professional phagocytic cells of bovine udder, polymorphonuclearneutrophils (PMN) and macrophages, comprise the first line of defense against invading mastitis pathogens. Most researchers now accept that the PMN is a key factor in the cows’ defense against intrama...

Systemic Activation of TLR3-Dependent TRIF Signaling Confers Host Defense against Gram-Negative Bacteria in the Intestine

Recognition of Gram-negative bacteria by toll-like receptor (TLR)4 induces MyD88 and TRIF mediated responses. We have shown that TRIF-dependent responses play an important role in intestinal defense against Gram-negative enteropathogens. In the current study, we examined underlying mechanisms of how systemic TRIF activation enhances intestinal immune defense against Gram-negative bacteria. Firs...

Host Immune Response to Influenza A Virus Infection

Influenza A viruses (IAVs) are contagious pathogens responsible for severe respiratory infection in humans and animals worldwide. Upon detection of IAV infection, host immune system aims to defend against and clear the viral infection. Innate immune system is comprised of physical barriers (mucus and collectins), various phagocytic cells, group of cytokines, interferons (IFNs), and IFN-stimulat...

Gene silencing without DNA. rna-mediated cross-protection between viruses

Previously, it was shown that the upper leaves of plants infected with nepoviruses and caulimoviruses are symptom free and contain reduced levels of virus. These leaves are said to be recovered. Recovery is associated with RNA-mediated cross-protection against secondary virus infection. Here, by analyzing plants infected with viruses that are quite distinct from the nepovirus or caulimovirus gr...
